C-erbB-2和Her-2人類基因是因其與逆轉(zhuǎn)錄病毒V—erbB-2相似而被發(fā)現(xiàn)。之后Coussens L及Schechte AL等對Neu,Her-2,C-erbB-2進(jìn)行測序及染色體定位分析,證明他們是同一基因。
人類該基因定位于染色體17q12,其編碼產(chǎn)物為185kD的跨膜精蛋白p185,由1255個(gè)氨基酸組成,720—987位屬于酪氨酸激酶區(qū)。Her-2/neu蛋白是具有酪氨酸蛋白激酶活性跨膜糖蛋白,是EGFR家族成員之一。
Her-2/neu蛋白由胞外的配體結(jié)合區(qū)、單鏈跨膜區(qū)及胞內(nèi)的蛋白酪氨酸激酶區(qū)三部分組成,由于目前尚未發(fā)現(xiàn)能與Her-2/neu蛋白直接結(jié)合的配體.其主要通過與家族中其他成員包括EGFR(HERl/erbBI),HER3/erbB3。HER4/erbB4形成異二聚體而與各自的配體結(jié)合。Her-2/neu蛋白常為異二聚體*伴侶,且活性常強(qiáng)于其它異二聚體。當(dāng)與配體結(jié)合后,主要通過引起受體二聚化及胞漿內(nèi)酪氨酸激酶區(qū)的自身磷酸化.激活酪氨酸激酶的活性。
現(xiàn)已證實(shí),HRE-2的過度表達(dá)可潛在激活EGFR的信號通路,同時(shí)可促進(jìn)EGFR介導(dǎo)的轉(zhuǎn)化和腫瘤的發(fā)生。HER-2/neu基因檢測試劑盒 熒光原位雜交法

Her-2/neu protein mediated signal transduction pathways include Ras/Raf/ mitogen activated protein kinase (MAPK) pathway, phosphatidylinositol.3 hydroxyl kinase (P13K) /Akt pathway, signal transduction and transcriptional activation (STAT) pathway, and PLC pathway. 2 the expression and influencing factors of Her-2/neu protein in gastric cancer. Her-2/neu protein is usually expressed only in the fetal stage, and it is expressed in a small number of tissues only after adult. However, it is overexpressed in many kinds of human tumors, such as breast cancer, ovarian cancer, lung adenocarcinoma, primary renal cell carcinoma and endometrial carcinoma. On the gastric cancer cells, the Her-2/neu protein is mainly located on the cell membrane, and a small amount is expressed in the cytoplasm. In gastric cancer, Her-2/neu gene amplification and RNA and protein overexpression were common, but not detected in all non gastric cancer tissues, indicating that Her-2/neu protein plays an important role in the occurrence, development and invasive metastasis of gastric cancer.
